The race will head around Goodwood airfield, into Lavant, down Pook Lane and Broyle Road, through the Festival Theatre grounds, finishing in Northgate car park.
College Lane will be closed both ways for the start of the race. Oaklands Way, Northgate Roundabout, Spitalfield Lane and Westhampnett roundabout will also be affected by the closure.
